Development of Neuron-Targeting Lipid Nanoparticles Functionalized with Cell-targeting Peptides for Enhanced mRNA Delivery to the Central Nervous System(CNS).

Abstract

Lipid nanoparticles (LNPs) represent a promising platform for mRNA-based therapy to CNS diseases. However, conventional LNPs exhibit limited efficiency in delivering mRNA therapeutics to neurons within the CNS, representing a significant challenge. To overcome this challenge, a strategy for developing neuron-targeting LNPs via conjugation with neuron-specific peptides is presented in this study. An M13 bacteriophage-based peptide phage display library was used for the discovery of neuron-specific peptides. Through cell-based biopanning on differentiated SH-SY5Y neuronal cell line, several peptide candidates were confirmed. Dye-conjugated peptides exhibited significantly increased uptake in both SH-SY5Y cell and murine CNS neurons. The evaluation of mRNA delivery efficiency using the selected peptide-LNPs will be performed both in vitro and in vivo. Overall, neuron-targeting peptide-LNPs hold potential as a sophisticated mRNA-delivery system for treating CNS-related diseases.
