Abstract
Current drug delivery systems have been developed, yet they suffer from limitations such as long-term toxicity from in vivo accumulation, immune responses, complex manufacturing processes, and low drug loading efficiency. Here, we introduced a simple coacervate that is biocompatible, easy to produce, and enables high-efficiency drug encapsulation. This system lies in using a senomorphic drug as the spacer moiety, allowing both the delivery vehicle itself and the loaded drug to exert rejuvenation effects. Senescence cell-targeting antibodies were conjugated to the surface to enhance cell specificity, enabling direct cytosolic delivery due to its small size; subsequently, the sticker moiety's disulfide bond is cleaved by overexpressed GSH in senescent cells. This process triggers dual release of the spacer senomorphic drug and the loaded additional drug. Our system is expected to maximize rejuvenation effects through multi-pathway targeting.
