Multilamellar Lipid Nanovaccines Incorporating Clumping Factor A and Dual TLR Adjuvants for Enhanced Anti–Staphylococcus aureus Immunity

Abstract

Methicillin-resistant Staphylococcus aureus (MRSA) poses a serious clinical challenge due to antibiotic resistance and the absence of effective vaccines. We present a biomimetic nanovaccine platform based on structurally reinforced multilamellar lipid encapsulating the fibrinogen-binding N23 domain of clumping factor A (ClfAN23) together with dual Toll-like receptor adjuvants. Incorporating ceramide ENP enables the formation of robust multilamellar architectures with markedly improved stability against lyophilization, repeated freeze-thaw cycles, and serum exposure. Optimized surface presentation of MPLA and CpG-Chol synergistically enhances dendritic cell maturation, co-stimulatory molecule expression, and proinflammatory cytokine secretion. In vivo immunization elicits strong antigen-specific humoral responses and Th1/Th17-skewed T cell immunity.
