Polydopamine-Mediated Hydrogel Adhesion Enables a PDMS 3D ALI Skin Scaffold

Abstract
3D skin scaffolds are crucial alternatives to animal testing, yet maintaining long-term structural integrity and user-friendly operation remains challenging. This study presents a human skin model based on 3D-printed polydimethylsiloxane (PDMS). By utilizing PDMS to perform air-liquid interface (ALI) culture, we simplified the culture procedure and designed the system to be suitable for large-scale production and automated processing. To address hydrogel contraction issues during long-term culture, polydopamine surface coating was applied to enhance adhesion between the scaffold and hydrogel. Through co-culture of endothelial cells, fibroblasts, and keratinocytes, we successfully recapitulated skin-like features including vascularized dermal structure and stratified epidermal structure. This model can be applied to drug evaluation and regenerative medicine.
