Development of a Serpentine-Patterned PCL Scaffold via Z-axis 3D Printing and Electrospinning

Abstract

Polycaprolactone (PCL) is a biocompatible, FDA-approved polymer widely used in tracheal reconstruction. Conventional PCL scaffolds adopt either solid cylindrical (body-type) or mesh-type designs. Body-type scaffolds offer mechanical strength but often experience coating delamination, while mesh designs improve coating retention via pore interlocking but lack durability. To address both issues, we developed a flexible tracheal scaffold by integrating Z-axis 3D printing with electrospun PCL nanofibers. This composite structure exhibited strong bonding without delamination, enabling reliable surface modification. Finite element analysis under simulated tracheal loading guided the use of a serpentine pattern for improved compliance. The scaffold demonstrated native-like flexibility, durability under repeated compression, and scalability beyond 6 cm. This platform presents a robust, manufacturable solution for repairing large-segment tracheal defects.
