Microfluidic-mediated tailored self-assembly of lipid nanoparticles for enhanced drug delivery

Abstract

Lipid-based nanoparticles are recognized as an essential component in drug delivery systems because of their biocompatibility and diverse applicability. It is essential to produce nanoparticles with a uniform structure for safe and effective delivery. We introduced a microfluidic chip that can control the size of nanoparticles and reduce variability. Lipid nanoparticles (LNPs) were manufactured using a microfluidic method and compared with liposomes manufactured using a thin film hydration method. The influence of flow rate and lipid concentration on the size distribution of LNPs was evaluated. LNPs with reduced size, lower PDI, and enhanced nucleic acid encapsulation were achieved through optimized the condition. The fabricated LNPs exhibited excellent cellular uptake and lower cytotoxicity, while ensuring efficient systemic clearance. This study suggests the potential of microfluidic LNPs as efficient nucleic acid delivery vehicles.
