Abstract
Burn wound healing remains a substantial clinical challenge owing to persistent inflammation and the absence of a pro-regenerative microenvironment. To address these obstacles, cell-derived extracellular matrix (cECM)-based hydrogels have emerged as promising candidates for modulating cellular behavior. In this study, we developed cECM-incorporated bioactive hydrogels (Gel-cECM) that exhibited enhanced mechanical properties while preserving ECM components, thereby facilitating cell-matrix interactions. Our findings revealed that Gel-cECM significantly promoted host cell infiltration, M2 macrophage polarization, angiogenesis, and collagen deposition in vivo. Importantly, Gel-cECM treatment expedited wound closure and supported the functional regeneration of skin appendages, including hair follicles, muscle fibers, and nerves. Collectively, these results demonstrate the therapeutic potential of Gel-cECM as an advanced platform for burn wound management and functional tissue regeneration.
