Development of an Efficient Levan Synthesis Process via Levansucrase Immobilization on Ni2+-His @ Fe3O4 NP Magnetic Nanoparticles

Abstract

Levan is a highly versatile biopolymer with significant potential in the pharmaceutical and biotechnological industries. However, its large-scale production is hindered by the enzymatic characteristics of levansucrase , which catalyzes the hydrolysis of synthesized levan upon the depletion of the sucrose substrate. This secondary reaction significantly reduces the final yield and complicates process control.
To address this, we developed Ni2+-His @ Fe3O4 magnetic nanoparitcles for efficient enzyme immobilization.
This system enables rapid enzyme recovery via magnetic force before levan degradation and ensures enzyme reusability. By evaluating the optimal pH, temperature, and thermal stability, we identified conditions that maximize enzymatic activity. These results provide a robust technical foundation for the commercial-scale production of levan by extending enzyme lifespan and enhancing overall yield.
