Multifunctional GQD-PDRN Nanocomposite-Loaded Hyaluronic Acid for Integrated Tissue Regeneration and NIR-responsive Photothermal Therapy in Diabetic Wound Healing

Abstract

Diabetic wounds are difficult to heal due to oxidative stress, infection, and impaired angiogenesis. To address these challenges, we developed a multifunctional hyaluronic acid (HA) hydrogel co-loaded with graphene quantum dots (GQDs) and polydeoxyribonucleotide (PDRN). GQDs, synthesized via a microwave-assisted method, exhibited strong reactive oxygen species (ROS) scavenging and near-infrared (NIR)-responsive photothermal properties. PDRN, known for stimulating tissue regeneration and angiogenesis, was embedded within the HA matrix. Upon NIR irradiation, GQD-mediated heating softened the hydrogel, enabling on-demand PDRN release and mild photothermal stimulation. This system enhanced fibroblast proliferation, supported angiogenesis, and accelerated diabetic wound healing in vitro with low cytotoxicity. These results suggest that the GQD-PDRN/HA hydrogel is a promising dressing platform integrating antioxidant, regenerative, and photothermal functions for diabetic wound treatment.
