Rationally designed bioactive milk-derived protein scaffolds enhanced new bone formation

Abstract

Casein contains bioactive peptides, including casein phosphopeptides and β-casochemotide-1, which induce macrophage chemotaxis and regulate bone homeostasis and fracture repair via cytokine secretion. We hypothesized that these multifunctional peptides could enhance bone regeneration. Accordingly, we developed a physically crosslinked milk-derived protein (MDP) scaffold composed of casein and polyvinyl alcohol using freeze–thaw cycles and further functionalized its surface with 3,4-dihydroxy-L-phenylalanine (DOPA). Both MDP and MDP-DOPA promoted macrophage migration of RAW 264.7 cells, indicating the activity of intrinsic biopeptides. In a mouse calvarial defect model, MDP-DOPA induced faster mineral deposition and higher bone density than other groups. Notably, MDP-DOPA recruited host M2 macrophages and mesenchymal stem cells around the scaffold, whereas MDP predominantly induced M1 macrophage accumulation.
