Super-Resolution Fluorescence Microscopy: From Technology to Applications

Abstract
Single-molecule localization microscopy (SMLM) enables nanoscale biological imaging with spatial resolutions down to ~20 nm but remains difficult to apply to complex systems such as thick tissues due to limited optical penetration, signal instability, and reconstruction accuracy. Our laboratory develops integrated engineering solutions to address these challenges, including a high refractive-index imaging buffer, a UV-curable resin sealant for long-term imaging, advanced point spread function modeling for accurate 3D localization, and deep learning-based background suppression. I will present representative biolobical applications, including nanoscale mapping of actin isoforms and modulation of cytoskeletal dynamics using graphene quantum dots in ocular surface disorders. Together, these studies demonstrate how advances in super-resolution microscopy enable quantitative biological insights and translational applications.
