Abstract
Messenger RNA (mRNA) therapeutics now extend beyond COVID-19 vaccines to protein replacement and immune modulation, but delivery is a central bottleneck because mRNA is unstable and must reach the cytosol. Lipid nanoparticles (LNPs) transfect efficiently, yet off-target distribution, innate immune activation, and organ toxicity drive interest in alternatives. Polymeric nanoparticles (PNPs), especially poly(β-amino ester) (PBAE) systems, offer biocompatibility, lower innate immunogenicity, and design flexibility. PBAEs allow systematic tuning via monomer choice and formulation to match specific applications. Recent work shows optimized PBAE nanoparticles can yield localized, prolonged mRNA expression after intramuscular injection while limiting systemic exposure and reducing liver-associated toxicity. Ongoing optimization positions PBAE-based PNPs as potentially safer, versatile carriers that broaden mRNA applications beyond current vaccine platforms.
