Abstract
Natural killer (NK) cells are key effectors of cancer immunosurveillance and can eliminate tumor cells via antibody-dependent cellular cytotoxicity (ADCC). However, heterogeneous tumor antigen expression limits NK-mediated ADCC. We developed a Universal Antibody (Univody) strategy enabling antigen-independent NK recognition by displaying IgG1 Fc fragments on cancer-cell surfaces. Univody is a fusion protein of the human IgG1 Fc fragment and a transmembrane domain, delivered as plasmid DNA (pUnivody) to achieve cell-surface localization and NK receptor engagement. A phenylboronic acid (PBA)-modified lipopolyplex (LPP) was used for delivery, enhancing uptake and transfection while reducing cytotoxicity. pUnivody@LPP-PBA promoted Fc display on tumor cells and activated NK cells in vitro. In triple-negative breast cancer and melanoma models in vivo, pUnivody@LPP-PBA inhibited tumor growth, supporting an antigen-independent immunotherapy with therapeutic potential for diverse cancers.
