Probing Extended Single Chain Morphologies in hc-PMMA via AFM and STORM

Abstract
Direct observation of individual polymer chains enables quantification of structural features beyond ensemble averages. Here, we investigate 1D hypo-crystal (hc) PMMA at the single-chain level by integrating complementary imaging modalities to link chain structure with anisotropic organization. Measurements combine AFM-based nanoscale imaging with STORM (stochastic optical reconstruction microscopy) as a super-resolution fluorescence readout under dilute-chain conditions, providing population-level statistics of chain dimensions and heterogeneity. AFM reveals elongated, fiber-like features in specific regions, suggesting that chains may adopt extended morphologies or form aligned polymer strands depending on the local environment. To probe extended conformations at higher resolution, we prepare hc-PMMA with end-tethered PMMA for STORM imaging. This framework provides quantitative descriptors to evaluate mechanisms promoting chain extension and anisotropic organization in hc-PMMA
