Abstract
Oral mucositis induced by chemotherapy or radiotherapy is a severe complication that can cause pain, malnutrition, bleeding, and interruption of cancer treatment. Conventional ointments, patches, and films show limited efficacy because they detach or are rapidly cleared in the moist, dynamic oral environment. Here, we develop an injectable, self-healing, and mucoadhesive starch-based hydrogel for protective and sustained exosome delivery. The hydrogel forms under mild conditions via dynamic Schiff base reactions between oxidized starch and amine-containing chitosan derivatives, helping preserve exosome integrity. Oxidized starch enables saliva-responsive degradation (amylase-triggered), promoting controlled release. Gallic acid functionalization provides wet adhesion while maintaining injectability and self-healing. This platform offers a promising approach for localized exosome delivery to treat oral mucositis.
