Abstract
Glomerular diseases are a major cause of chronic kidney disease, yet the lack of physiologically relevant in vitro models limits long-term culture and effective drug screening at the glomerular level. To address these limitations, we developed a vascularized glomerulus-on-a-chip platform for sustained ex vivo culture of isolated glomeruli.
In this system, intact glomeruli are integrated into a microfluidic device where microvasculature is formed within a collagen-based hydrogel and connected to external reservoirs. Gravity-driven flow generated by hydrostatic pressure differences enables continuous media exchange and vascular access without external pumps. This platform supports prolonged glomerulus maintenance under perfused conditions and provides a practical model for glomerulus-level drug screening. Overall, this approach offers a simplified and physiologically relevant alternative for kidney disease research and preclinical drug evaluation.
