Proteolytically Robust Peptoid-Peptide Hydrogels for Healthcare Applications

Abstract

Peptide-based supramolecular hydrogels are attractive for cargo encapsulation but often suffer from instability and rapid degradation. Here, we introduce peptide–peptoid hybrid hydrogels as a strategy to enhance structural robustness while retaining supramolecular assembly. A peptoid-grafted molecule, FmocFFG-Nsar4, was synthesized and co-assembled with FmocFF to systematically vary peptoid content within the network. Hydrogels formed via solvent-switch self-assembly and were characterized structurally and mechanically to evaluate gelation, microarchitecture, viscoelasticity, and stress resistance. Peptoid content strongly affected assembly, revealing a critical composition window. Rheology showed stable viscoelastic behavior with G' > G" and notable self-healing. These features produced enhanced stability relative to peptide-only gels, highlighting hybrid peptide–peptoid hydrogels as tunable systems for diverse applications.
