Indocyanine Green J-aggregate Nanosuspension with High Loading Efficiency for Photothermal Therapy

Abstract
Indocyanine green (ICG) has been widely investigated for applications in photothermal therapy (PTT). J-aggregated ICG (J-ICG) has been employed to enhance molecular stability and quantum efficiency. However, J-ICG are prone to dissociation in solution when combined with other materials during particle formation. In this study, a PLL@J-ICG nanosuspension was successfully prepared by wet media milling of J-ICG with Poly-L-lysine (PLL) in methanol, enabling the preservation of the J-aggregate structure through electrostatic interactions between PLL and J-ICG. The resulting nanoparticles exhibited an average particle size of 319.7±101.0 nm, and the milling-based approach achieved higher loading amount compared to conventional bottom-up methods. in vitro photothermal conversion studies demonstrated that PLL@J-ICG exhibited photothermal conversion efficiency comparable to that of free J-ICG, indicating its strong potential as a therapeutic and imaging agent for PTT applications.
