GelMA-Based Electroconductive Hydrogel Nanocomposite for Accelerating Diabetic Wound Regeneration

Abstract

Diabetic wounds delay the recovery cycle and lead to ulcers and amputation. Electrostimulative therapy (EST) has been investigated to promote wound healing, but current EST materials face challenges of limited biocompatibility and unsuitable properties. This study developed an EST hydrogel composed of gelatin methacryloyl (GelMA), MXene, and DNA. GelMA offers biocompatibility and tunable mechanical properties. Incorporation of MXene enhances electrical conductivity and strength, but may induce cytotoxicity by oxidation. To address this, MXene was coated with polydopamine (PDA) and DNA to improve biocompatibility and tissue regenerativity. FT-IR, XPS, and DLS confirmed surface modification. The composite hydrogel showed improved mechanical and electrical properties. In vitro results demonstrated reduced cytotoxicity and enhanced cell proliferation by EST. These findings suggest the hydrogel composite as a promising platform for skin tissue regeneration by EST.
