Nettle-Inspired Microneedles for Localized Skin Cancer Therapy and Tissue Regeneration

Abstract

Surgical treatment for skin cancer remains the primary method for managing malignant tumors, but recurrence from damaged tissues and residual tumors remains a challenge. To address this, we developed nettle-inspired microneedles that promote tissue regeneration and localized tumor therapy. Fabricated using digital 3D printing and soft lithography, the microneedles were enhanced by biomineralizing salmon sperm DNA with silica, improving insertion and drug delivery efficiency. Loaded with chlorin e6, a photosensitizer that generates reactive oxygen species under near-infrared irradiation, they induced melanoma cell apoptosis. Beyond tumor ablation, the microneedles triggered anti-inflammatory responses, collagen deposition, and re-epithelialization, accelerating skin regeneration. This multifunctional platform combines photodynamic therapy and regenerative medicine, offering a promising strategy for post-surgical cancer care and biomedical applications.
