A Multimeric Polyethylenimine Carriers for mRNA Delivery

Abstract
Messenger RNA (mRNA) is a promising therapeutic platform capable of rapid and transient protein expression without genomic integration. However, its application is limited by low stability and inefficient cellular delivery. In this study, low-molecular-weight PgP polymers (0.8 kDa and 2 kDa) were developed as non-viral mRNA carriers. PgP polymers were synthesized via Michael addition and amidation reactions and characterized by ^1H NMR and FT-IR spectroscopy. PgP showed buffering capacity comparable to PEI 25 kDa and efficient mRNA condensation into stable, positively charged nanocomplexes. Confocal microscopy confirmed efficient cellular uptake and cytosolic localization in NIH3T3 cells. Transfection experiments showed luciferase expression levels comparable to PEI 25 kDa while maintaining over 90% cell viability. PgP 2 kDa exhibited higher transfection efficiency, whereas PgP 0.8 kDa showed superior biocompatibility, indicating its potential as a safe non-viral mRNA delivery system.
