Abstract

Chronic foot ulcers are a serious complication of diabetes, requiring wound dressings that enable rapid hemostasis and support tissue regeneration. In this study, we developed a 3D-printed hydrogel patch for diabetic wound treatment. A biocompatible polymer matrix was combined with nanoclay and DNA to introduce bioactivity and reinforce the hydrogel network. The nanoclay served as a carrier for DNA and enhanced hydrogel integrity, resulting in improved mechanical stability and printability. The bioink was fabricated into wound-conforming patches using a 3D bioprinter. Cytocompatibility was evaluated using fibroblasts. Hemostatic performance was assessed using whole blood, demonstrating rapid clot formation and low hemolytic activity. In vivo diabetic wound models further showed accelerated hemostasis and enhanced wound closure. Overall, this 3D-printed functional hydrogel patch shows strong potential as a hemostatic and regenerative platform for diabetic wound healing.
