3D-Printed Biofunctional GelMA-Based Layered Scaffolds for Cartilage Defect Repair

Abstract

Cartilage defects, caused by trauma or degeneration, have limited self-healing capacity due to the avascular and aneural nature of cartilage tissue. In this study, we developed a customized 3D-printed hydrogel scaffold based on gelatin methacryloyl (GelMA) that mimics the zonal architecture of native cartilage and provides a biocompatible platform for tissue regeneration. To enhance mechanical strength and enable sustained delivery of bioactive molecules, a nanoclay with electrostatic interaction potential was incorporated. A polyamine known to promote chondrogenesis, immune modulation, and cell viability was combined with the nanoclay for controlled release. This composite configuration allowed the scaffold to maintain structural integrity. The scaffold's mechanical properties, release behavior, and biological performance were evaluated using adipose-derived stem cells. This integrated approach presents a promising strategy for cartilage repair.
