Angiogenesis in Mesenchymal Stem Cell/Endothelial Cell Spheroids Incubated under Simulated Microgravity

Abstract

The engineering of three-dimensional (3D) artificial tissues and organs has emerged as a highly promising approach in regenerative medicine. However, the size of engineered tissues is still largely limited to sub-millimeter dimensions due to insufficient vascularization. While co-culture strategies involving mesenchymal stem cells (MSCs) and endothelial cells (ECs) have been explored to facilitate vascular formation, achieving rapid and robust angiogenesis remains challenging. Previous studies have suggested that microgravity (μg) conditions can enhance angiogenic responses through cytoskeletal remodeling. In this study, we prepared MSC/EC spheroids and incubated a simulated microgravity platform based on neutral buoyancy to create vascular structure in the 3D constructs. We determined optimal ratio of MSCs and ECs for angiogenesis under simulated μg. Angiogenic-related genes under simulated μg were compared with normal gravity through qRT-PCR and immunofluorescence staining.
