Fabrication and Characterization of Thermogel Film Formulations Incorporating Stem Cell–Derived Conditioned Medium

Abstract

Mesenchymal stem cell–derived conditioned medium (MSC-CM) has gained attention as a cell-free therapeutic approach due to bioactive factors secreted through paracrine effects, although its direct application is limited by rapid diffusion from the target site. In this study, an OGC-CM thermogel film was developed by integrating MSC-CM into an octanoyl glycol chitosan–based thermogel formulation. Due to its temperature-responsive behavior, the thermogel can be easily processed in the sol state and maintained in the gel state under physiological conditions. The OGC-CM thermogel films exhibited rapid hydration and high water uptake exceeding 2000%, and tissue adhesion tests confirmed stable interfacial attachment under wet conditions. In vitro evaluations suggested a favorable biological environment relevant to wound healing and infection-related conditions, indicating that OGC-CM thermogel films represent a promising wound dressing platform.
