Preparation and Characterization of Injectable OGC/ECM Hybrid Hydrogels for Tissue Engineering

Abstract

Thermogel undergo sol–gel transitions near physiological temperature, enabling rapid gelation after injection. Extracellular matrix (ECM) provides tissue-specific biochemical cues. In this study, an injectable OGC/ECM hybrid hydrogel was developed by integrating cartilage derived extracellular matrix (cECM) with octanoyl glycol chitosan (OGC) followed by the incorporation of dibenzaldehyde-terminated polyethylene glycol (DP) as a dynamic crosslinker. Gelation of OGC was driven by hydrophobic interactions while the incorporation of cECM is expected to enhance the bioactivity. Further, DP reinforced the OGC/ECM hydrogel through dynamic Schiff base crosslinking, improving its mechanical strength. The mechanical properties of the hydrogels were evaluated using rheological analyses. Overall, the injectable OGC/ECM hybrid hydrogel demonstrates potential as a scaffold material for tissue engineering due to its minimally invasive injectability and rapid in-situ gelation.
