Stem cell-derived extracellular vesicles-loaded hyaluronic acid hydrogel for skin anti-aging

Abstract
With the rising demand for effective anti-aging interventions, hyaluronic acid–based hydrogels (Hyal-Gels) serve as a foundational dermal filler. Here, we developed EVHyal-Gels by loading adipose-derived stem cell (ADSC) extracellular vesicles (EVs) into Hyal-Gels to enhance dermal regeneration. This approach leverages the ability of ADSCs to promote a CD301b-expressing macrophage phenotype, which drives fibroblast proliferation and collagen synthesis. In a mouse model, EVHyal-Gels extended the biological half-life of EVs from 1.37 to 3.75 days. We identified key miRNAs, specifically let-7b-5p and miR-24-3p, as the primary mediators of the macrophage transition to the CD301b+ phenotype. Histological analysis confirmed that EVHyal-Gels induced a 2.4-fold increase in dermal collagen area compared to standard Hyal-Gels. These findings demonstrate that EVHyal-Gels effectively improve the dermal microenvironment, offering a potent therapeutic strategy for skin rejuvenation.
