Abstract
Inspired by natural melanogenesis, we developed a tyrosinase-mediated coating strategy to form eumelanin films functionalized with N-oxide molecules. Amine-terminated N-oxide compounds were synthesized via Boc protection and subsequent deprotection, and were grafted onto eumelanin-coated surfaces via Michael addition and Schiff-base reactions. The formation of eumelanin films and the successful incorporation of N-oxide species were systematically characterized by contact angle goniometry, ellipsometry, X-ray photoelectron spectroscopy, and atomic force microscopy. This coating approach was successfully applied to a wide range of substrates, demonstrating its versatility. The N-oxide–functionalized eumelanin coatings effectively reduced adhesion of the Gram-negative Escherichia coli and maintained structural stability under both physiological and strongly acidic conditions, indicating strong potential for antibacterial surface engineering applications.
