Dual NIR-Responsive Hyaluronate - Black Phosphorus - Upconversion Nanoparticle Complex for Image-Guided Skin Cancer Phototherapy

Abstract

Non-invasive, image-guided phototherapy remains challenging due to limited tissue penetration and lack of depth-resolved control. To address this, we developed a hyaluronate -black phosphorus - upconversion nanoparticle (HA-BP-UCNP) complex enabling multimodal theranostics of skin cancer, including photoacoustic (PA) imaging, photodynamic therapy (PDT), and photothermal therapy (PTT). Hyaluronic acid enables enhanced skin permeation and tumor accumulation of the nanocomplex. Under 980 nm NIR light, the UCNP emits UV-blue light, which activates the BP component for reactive oxygen species (ROS) generation for PDT. Simultaneously, 808 nm NIR light irradiation induces localized photothermal effects for effective tumor ablation. Furthermore, the intrinsic PA signal from BP enables non-invasive visualization of the nanocomplex distribution and tumor localization in vivo. This nanoplatform demonstrates strong potential for non-invasive, image-guided, and synergistic treatment of skin cancer.
