Multidimensional Polymer Design Framework for Tissue Processing and Interrogation

Abstract
Deciphering biological physiology and pathology requires high-clarity, high-resolution three-dimensional maps. Hydrogel-based tissue clearing enables volumetric imaging and phenotyping while preserving native structural and molecular features. Despite diverse tissue clearing hydrogels with tailored physicochemical properties, a unified polymeric materials science and engineering perspective remains lacking. Here, we present a multidimensional framework for designing high-performance tissue clearing hydrogels, integrating (i) chemorheological analysis, (ii) swelling behavior, (iii) porosity evolution, (iv) molecular probe transport, (v) mechanical performance, and (vi) tolerance to thermo-chemical perturbations. The interplay among these parameters governs platform functionality and bioengineering workflow efficiency, enabling systematic development of next-generation tissue clearing hydrogels for advanced, multi-omics interrogation.
