Injectable Nanofiber System Enabling Prolonged Tumor Retention of Immune-Activating Complexes for Cancer Immunotherapy

The efficacy of cancer immunotherapies is constrained by immunosuppressive tumor microenvironments and immune tolerance mechanisms. This study developed an injectable nanofiber system loaded with immune-activating complexes. The surface of biomaterial-based nanofibers was modified with polymeric coating to enable electrostatic attachment of immune-activating complexes, additionally exhibiting the effect of promoting immune cell proliferation. Electron microscopy confirmed complex immobilization and cytotoxicity assessment established biocompatibility. Immune cells showed significantly increased pro-inflammatory cytokines and immune activation biomarkers. In vivo tumor model experiments using fluorescently-labeled complexes demonstrated that the nanofiber carrier enabled prolonged tumor retention, thereby inducing sustained immune activation. The platform presents a promising strategy for cancer immunotherapy.
