mRNA-based therapeutics have emerged as a versatile platform for cancer treatment owing to their high expression efficiency and programmability. However, most current strategies rely on expressing cytotoxic or immune-modulating proteins that act through specific molecular targets. Here, we describe an mRNA design encoding intracellular self-assembling proteins capable of forming multiple aggregates in cancer cells, leading to pronounced changes in intracellular protein organization. Such aggregation suggests a physically driven mechanism that may disrupt normal intracellular structure and function. Because this strategy is independent of specific molecular targets, it may offer an advantage in addressing cancer cell heterogeneity. Together, these findings suggest intracellular protein aggregation as a promising physical approach for mRNA-based cancer therapy.
