Recapitulating the tumor microenvironment (TME) remains challenging due to the roles of extracellular matrix (ECM) mechanics and cellular organization. Here, we present a cancer-on-a-chip platform based on microfluidic-fabricated ECM microgels that integrates tumor spheroids with stromal components in a three-dimensional microenvironment.
Uniform droplet-based ECM microgels were generated using a T-junction microfluidic system, enabling reproducible fabrication suitable for high-throughput applications. The microgels supported dense cellular encapsulation while maintaining structural stability. A layered microgel architecture was further developed to model brain tumor–specific microenvironments, providing a scalable and biomimetic platform for cancer modeling and drug screening
