Angiogenic Factor-loaded PVA-Alginate Microneedle Enhances Perfusion Recovery in a Mouse Hindlimb Ischemia Model

Effective revascularization in ischemic tissue requires localized delivery of pro-revascularization therapeutics. We developed solvent-cast, air-dried PVA–alginate microneedle (MN) arrays for localized protein delivery and observed improved perfusion recovery in a mouse hindlimb ischemia model via angiopoietin-1 (Ang-1) loading. By optimizing the polymer formulation, we obtained sharp tips with reduced demolding deformation (optical/SEM). Ang-1 loading and releasing profiles were quantified under physiological conditions. In vitro assays showed no cytotoxicity, and compression testing supported handling and penetration. Doppler imaging showed improved perfusion recovery with Ang-1–loaded MNs versus controls, while polymer-only MNs also improved. Overall, air-dried PVA–alginate MNs provide a practical platform for ischemic hindlimb therapy with an additive benefit from Ang-1, supporting translation to other ischemic tissue applications.
