Human mesenchymal stem cells (hMSCs) hold great potential for regenerative medicine, yet their variable in vivo behavior limits clinical translation. This study presents biocompatible calcium phosphate (CaP) nanoparticles stabilized with catechol-modified hyaluronic acid (dopa-HA) as a gene delivery platform to enhance hMSC osteogenic differentiation.
By leveraging the specific HA-CD44 receptor interaction, dopa-HA/CaP nanoparticles significantly improve transfection efficiency compared to branched polyethyleneimine (bPEI, 25 kDa) without cytotoxicity. Furthermore, co-delivery of BMP-2 pDNA and miRNA-148b through this system increases BMP-2 expression while downregulating Noggin, a natural inhibitor of BMP-driven osteogenesis. This dual-action strategy effectively bypasses biological bottlenecks, providing a robust and safe methodology for advanced bone tissue engineering.
