Enhanced Synthesis of Circular RNA via 1-nt Gapped Splint Ligation

In recent decades, there has been an uprising demand for sustainable RNA therapeutics for many diseases. The successful commercialization of mRNA vaccines and therapeutics has propelled RNA-based modalities to the forefront of modern medicine. However, most current mRNA therapeutics utilize a linear topology, which renders them susceptible to exonuclease-mediated degradation, thereby limiting their intracellular half-life and stability. To overcome these limitations, circular RNA (circRNA) has emerged as a promising alternative due to its inherent resistance to exonucleases. In this study, we propose an enhanced circularization strategy that introduces a 1-nucleotide (1nt) gap within the linear RNA substrate. By utilizing a DNA splint-mediated ligation approach with T4 RNA ligase2. And with this specific gap design, we achieved significantly higher circularization efficiency compared to conventional non-gap ligation methods
