Chemoimmunotherapy, which combines cytotoxic anticancer drugs with immunomodulators, has emerged as a promising strategy for the treatment of metastatic cancer by simultaneously inducing tumor cell death and stimulating cancer-specific immune responses. In this study, we developed a dissolving microneedle (MN) platform based on an amphiphilic triblock copolymer to enable efficient delivery of therapeutic agents to cancer-draining lymph nodes.
The MN system forms nanomicelles (PTX/R848@NMC) that co-encapsulate the poorly water-soluble chemotherapeutic agent paclitaxel (PTX) and the immune adjuvant resiquimod (R848). This lymph node–targeted delivery strategy induces metastatic cancer cell death while promoting dendritic cell (DC) activation and maturation, thereby enhancing antitumor immune responses. Collectively, our results highlight the potential of this MN-based drug delivery platform as an effective chemoimmunotherapeutic strategy for the treatment of metastatic cancer.
