Stem cell–based wound healing is limited by insufficient recruitment and retention of therapeutic cells at injury sites. In this study, we developed a click-crosslinked hyaluronic acid (Cx-HA) dressing incorporating an SDF-1–mimetic peptide (SMP) to promote in situ regeneration. The dressing enabled sustained release of SMP, providing a stable chemotactic signal at the wound site. In vivo, SMP-loaded Cx-HA accelerated wound closure and promoted long-term activation of both injected human mesenchymal stem cells (hMSCs) and host-derived murine mesenchymal stem cells (mMSCs), contributing to tissue remodeling and reduced scar formation through the stepwise regulation of chemotaxis, cell adhesion, and angiogenesis. Furthermore, SMP exhibited superior stability and lower production cost compared with recombinant SDF-1, highlighting its potential for clinical translation.
