The development of multifunctional nanotherapeutics offers a promising strategy to address key limitations of conventional cancer therapy. Herein, we present a multi-shell upconversion nanoparticle (UCNP)–based nanoplatform designed for triple-synergistic combination therapy integrating three different kinds of treatments. The UCNP core exhibits enhanced red emission under 808 nm near-infrared (NIR) irradiation and is functionalized with folic acid–polyethylene glycol (FPEG) to promote tumor-targeted delivery. An oxidoreductase enzyme, an iron-based organometallic compound, and a photosensitizer are conjugated onto the FPEG-modified UCNPs. In vitro studies demonstrate that nanoplatform exhibits preferential cellular uptake in HeLa cells, induces elevated intracellular reactive oxygen species (ROS) levels, and achieves synergistic cytotoxicity compared with single- or dual-treatments.
