Composite drug systems provide a platform for regulating structural stability and release behavior through controlled crystallization. In this study, N-acetylcysteine (NAC)–catechin composites were prepared via reverse antisolvent crystallization, and the effects of polymer addition, injection sequence, and crystallization temperature (15 °C and 40 °C) were examined. A 50:50 composition was confirmed by NMR regardless of polymer presence, indicating successful composite formation. FE-SEM analysis showed that, without polymer, the drug injected first tended to cover the particle surface at both temperatures, resulting in sequence-dependent morphology. In contrast, PLA addition promoted composite-like structures at elevated temperature. HPLC results revealed rapid release within 80–100 min for polymer-free samples, whereas PLA-containing samples exhibited sustained release extending up to 1 day. These findings demonstrate that polymer addition and temperature cooperatively regulate crystallization pathways and long-term release behavior.
