Hydrogel-based scaffolds have been extensively investigated in tissue engineering to promote tissue regeneration; however, recreating the physiological microenvironment with appropriate biological and mechanical cues remains a significant challenge. In this study, we developed a mechanically graded hydrogel incorporating L929 mouse fibroblast spheroids to mimic the native dermal stiffness microenvironment and evaluate its effect on full-thickness skin regeneration. The gradient hydrogel was fabricated via photocrosslinking, systematically characterized in vitro, and assessed for wound healing applications. The prepared gradient hydrogel exhibited accelerated wound closure and reduced scar formation compared to uniform hydrogels. Collectively, this biocompatible gradient hydrogel, which reproduces the physical stiffness gradients of skin tissue, represents a promising scaffold for effective regeneration of full-thickness skin defects.
