In this study, we present a single-particle SEIRAS platform based on single-crystalline silver microcubes to address the low throughput and high cost issues of conventional nanolithography-based fabrication. The chemically synthesized silver microcubes can be produced in large quantities and offer significantly reduced Ohmic losses compared with polycrystalline nanoantennas due to their single-crystalline structure. In particular, the cube geometry with sizes of 1.6–2.5 μm resonates well in the mid-infrared wavelength range, generating strong dipolar plasmon modes. The flat facets and sharp edges of the microcubes are highly favorable for local electric field enhancement, enabling sensitive detection of PDMS monolayers and BSA protein vibrational modes at the single-particle level. As a lithography-free, high-performance plasmonic sensor, this platform demonstrates strong potential for advanced applications in biosensing and clinical spectroscopy.
