High-Loading Polyhydroxyalkanoate (PHA) Nanocarriers for Enhanced Skin Delivery

Abstract

Polyhydroxyalkanoates (PHAs) are biopolyesters produced by microorganisms such as bacteria and algae. Their biodegradability and biocompatibility have driven growing interest in PHA-based nanomaterials for biomedical applications, particularly as bio-adhesives [1]. In this work, we developed PHA nanoparticles as versatile carriers for skin-targeted delivery systems, designed to transport active compounds such as ceramide and niacinamide. Using multiple emulsification methods in both hydrophilic and hydrophobic solvent systems, we obtained nanoparticles with precisely controlled sizes of 10–300 nm and narrow distributions. These particles achieved high encapsulation efficiencies for both lipophilic and hydrophilic active ingredients, enhancing their stability and bioavailability. Furthermore, the synergistic bioactive effects of PHAs and encapsulated active ingredients offer intrinsic skin benefits, distinguishing them from conventional delivery systems and underscoring their potential for cosmetic and biomedical applications.
