Abstract

Extracellular vesicles have gained significant attention in cell therapy and regenerative medicine for their biocompatibility and ability to transfer therapeutic cargos, including growth factors, nucleic acid, cytokines, and miRNAs, which regulate cellular behavior, and essential biological processes such as cell regeneration and differentiation. Although EVs have been effectively utilized to enhance myogenic, chondrogenic, and osteogenic differentiation of mesenchymal stem cells (MSCs), their potential to directly influence the differentiation of pluripotent stem cells (PSCs) for therapeutic applications remains underexplored. PSCs, characterized by their high proliferation rates, often exhibit inefficient uptake of foreign materials, including EVs. In this study, we present a novel strategy to enhance EV uptake in PSCs by overexpressing a fusion protein comprising fibronectin’s RGD and HepII fragments along with LAMP2B to promote FN display on EV surfaces.
