Oxygen- and calcium ion-releasing gelatin cryogels for enhanced hemostasis and skin wound healing

Abstract

Conventional hemostatic materials, such as gauze and bandages, are associated with delayed hemostasis and necessitate removal. To address these limitations, cryogels have garnered attention as hemostatic dressings due to their water absorption capacity and extended storage stability. Herein, we develop oxygen (O2)-generating cryogels (Oxycryogels) for use as hemostatic dressings via calcium peroxide (CaO2)-mediated disulfide bond formation and lyophilization. The Oxycryogels demonstrate improved mechanical properties and highly interconnected macroporous structures. They facilitate blood absorption and hemostasis by releasing Ca2+ from CaO2, both in vitro and in vivo. Moreover, O2-releasing kinetics are modulated via catalase treatment. Our Oxycryogels promote wound healing and angiogenesis by their porosity and oxygen release capabilities in vivo. Consequently, Oxycryogels represent a promising candidate for hemostat dressings, owing to their hemostatic and angiogenic effects.
