Advanced Osmium Redox Polymer Biosensors with Improved Enzyme Stability and Electron Transfer

Abstract

Self-monitoring of blood glucose (SMBG) via finger-prick sampling causes discomfort and offers only momentary glucose values. To overcome this, continuous glucose monitoring (CGM) biosensors have been developed, with enzyme-based systems offering high selectivity and sensitivity. While mediators like osmium (Os) complexes improve electrode stability, CGM still faces challenges in accuracy and long-term enzyme stability. To address this, we enhanced biosensor performance through Os complex modification and linker integration. Among tested complexes, the one with dmbpy ligands showed the lowest redox potential, improving electron transfer. Increasing the coordination number from four to five improved water solubility by preventing cross-linking. Ethylamine was introduced as a linker to covalently bind the polymer to the enzyme, strengthening interactions and enhancing current stability.
