Numerical evaluation of mixing performance in microfluidic systems for lipid-based nanocarrier production

Abstract

Microfluidic mixing plays a key role in liposome and lipid nanoparticle (LNP) synthesis, where solvent exchange drives self-assembly. While various mixer designs enable size control, the relationship between mixing behavior and nanocarrier formation remains unclear. In this study, we numerically evaluate three microfluidic mixers—Hydrodynamic Flow Focusing (HFF), Toroidal Mixer, and Microfluidic Vortex Focusing (MVF)—by quantifying the degree of mixing based on solute concentration profiles. Simulation results are correlated with experimental liposome synthesis to identify mixing characteristics that promote uniform, controllable nanocarrier formation. Based on these findings, we propose and demonstrate optimized microfluidic mixer design through liposomes and LNPs synthesis under defined flow conditions. This work offers a practical framework that links numerical analysis with experimental outcomes in the development of microfluidic platforms for lipid-based nanocarrier production.
