Cationic Copolymer Interfaces on Nanoneedles via Initiated Chemical Vapor Deposition for Intracellular Gene Delivery

Abstract

High-aspect-ratio nanoneedles enable direct intracellular delivery of therapeutic biomolecules by physically penetrating the cell membrane. To achieve high delivery efficiency, the surface properties and biocompatibility of nanoneedles must be carefully tuned. In this study, we employed initiated chemical vapor deposition (iCVD) to functionalize silicon nanoneedles with a copolymer of glycidyl methacrylate (GMA) and dimethylaminoethyl methacrylate (DMAEMA). This solvent-free, low-temperature process preserves nanoneedle structure while allowing precise surface functionalization. Quaternary ammonium groups from DMAEMA promote electrostatic binding with negatively charged biomolecules, and epoxy groups from GMA enhance hydrophilicity and cell adhesion. The polymer-coated nanoneedles improved cell adhesion and viability while enabling efficient intracellular delivery. Our iCVD-based copolymer functionalization strategy demonstrates improved performance for nanoneedle-mediated delivery.
