Long-Term Stabilization of Nucleic Acids Using Ionic Liquids

Abstract

Among various types of nucleic acids, mRNA has emerged as a leading next-generation vaccine material, particularly highlighted during the COVID-19 pandemic. Despite its potential, mRNA is inherently unstable in aqueous environments due to hydrolysis. Accordingly, mRNA vaccines can only be stored for about 12 hours at room temperature and up to 6 months at cryogenic temperatures due to their limited stability. Furthermore, repeated freeze-thaw cycles can compromise nucleic acid stability. In this study, we introduce a novel stabilization strategy utilizing an ionic liquid. High-concentration cations in the ionic liquid interacts with the phosphate backbone of nucleic acids at hydrolysis-prone sites, thereby protecting them from structural degradation. This approach holds promise for enabling the long-term preservation of nucleic acids at ambient temperatures while maintaining their structural integrity and biological activity.
