Antioxidation-functionalized Hyaluronic Acid Conjugates for Oxidative Stress-Mitigating Cancer Therapy

Abstract

Hyaluronic acid (HA) is a biocompatible and biodegradable glycosaminoglycan widely used in medical applications. It is especially studied in cancer-targeting therapies due to its ability to bind to CD44 receptors, which are overexpressed in various tumors. Elevated levels of reactive oxygen species (ROS) are commonly observed in tumor environments, contributing to various aspects of cancer development and progression. Polyphenols, plant-derived antioxidants with multiple phenolic groups, have been widely studied for their biological activities. Among them, Tannic acid (TA), which contains ~25 phenolic hydroxyl groups, is known for its strong antioxidant, antibacterial, and anti-inflammatory properties. In this study, we synthesized HATA conjugates by chemically linking TA to HA using epichlorohydrin. The conjugates showed concentration-dependent free radical scavenging ability, unlike native HA, indicating its potential as an antioxidant material for tumor-targeted applications.
